A gene on chromosome 12q24.31 that encodes a SET domain-containing protein-lysine N-methyltransferase that monomethylates Lys-20 of histone H4 (H4K20me1). H4K20me1 is enriched during mitosis and is a specific tag for epigenetic transcriptional repression, found primarily in euchromatin regions where it plays a central role in silencing euchromatic genes. SETD8 is required for cell proliferation and helps maintain proper higher order structure of DNA during mitosis related to chromosome condensation and cytokinesis. It also mediates monomethylation of p53/TP53 at Lys-382, repressing p53/TP53-target genes.
