au·to·he·mol·y·sis test

when sterile defibrinated blood is incubated at 37°C, normal red blood cells hemolyze slowly; cells with membrane or metabolic defects do so to a greater extent. Test will detect spontaneous hemolysis of red blood cells.

autohemolysis test

An assay that measures the amount of spontaneous haemolysis that occurs after incubation of defibrinated blood at 37ºC after 48 hours. The results obtained are compared with those from identical blood enriched with glucose or ATP before incubation, allowing the stratification of haemolytic anaemias (glucose-6-phosphate dehydrogenase, pyruvate kinase, and others) and hereditary spherocytosis into 3 abnormal patterns. Because of the unwieldiness of the test, it is rarely used.
Indications
Diagnosis hereditary spherocytosis, spontaneous haemolysis, RBC enzyme deficiencies and haemolytic anaemias.
