IL10
A gene on chromosome 1q31-q32 that encodes interleukin-10, a cytokine produced primarily by monocytes, but also by lymphocytes, which inhibits the synthesis of a number of cytokines (including IFN-gamma, IL-2, IL-3, TNF and GM-CSF) produced by activated macrophages and helper T cells. IL10 has pleiotropic effects in immunoregulation and inflammation. It downregulates expression of Th1 cytokines, MHC class-II antigens and co-stimulatory molecules on macrophages, and it enhances B cell survival, proliferation and antibody production. It can block NF-kappa B activity, and is involved in regulating the JAK-STAT signalling pathway.Molecular pathology
IL10 mutations are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.
